目的研究小鼠脾脏间充质干细胞(Sp-MSCs)对T淋巴细胞增殖与活化的调节作用。方法采用组织块法从小鼠脾脏中分离培养出Sp-MSCs,并做多向诱导分化实验。分离小鼠T淋巴细胞,开展淋巴细胞转化试验和混合淋巴细胞反应,分别检测Sp-MSCs对于非特异抗原和同种异体抗原活化的T淋巴细胞的影响。以CFSE流式法检测Sp-MSCs对活化的T淋巴细胞增殖的影响。采用定量PCR分析Sp-MSCs对T淋巴细胞表达的炎性细胞因子的影响。结果流式检测结果显示Sp-MSs C可以抑制非特异抗原和同种异体抗原引起的T淋巴细胞增殖。Sp-MSCs能够有效抑制淋巴细胞转化试验和混合淋巴细胞反应引起的细胞活化。进一步分析发现,Sp-MSCs抑制T淋巴细胞表达干扰素γ,肿瘤坏死因子α和白细胞介素17A。结论 Sp-MSCs能够抑制T淋巴细胞增殖与活化,抑制T淋巴细胞表达多种炎性细胞因子。
Objective To investigate the regulatory effect of mouse splenic MSCs (Sp-MSCs) on the proliferation and activation of T lymphocytes. Methods Sp-MSCs were isolated and cultured from mouse spleen by tissue block method. T lymphocytes were isolated from mice, lymphocyte transformation test and mixed lymphocyte reaction were performed to detect the effect of Sp-MSCs on T lymphocytes activated by non-specific antigen and alloantigen. The effects of Sp-MSCs on the proliferation of activated T lymphocytes were detected by CFSE flow cytometry. The effect of Sp-MSCs on inflammatory cytokines expressed by T lymphocytes was analyzed by quantitative PCR. Results The results of flow cytometry showed that Sp-MSs C could inhibit the proliferation of T lymphocytes induced by non-specific antigen and alloantigen. Sp-MSCs can effectively inhibit lymphocyte transformation test and mixed lymphocyte reaction-induced cell activation. Further analysis found that Sp-MSCs inhibited T lymphocytes expressing interferon gamma, tumor necrosis factor alpha and interleukin 17A. Conclusion Sp-MSCs can inhibit the proliferation and activation of T lymphocytes and inhibit the expression of many inflammatory cytokines in T lymphocytes.
