目的研究白藜芦醇对大鼠肝癌细胞CBRH7919凋亡的诱导作用。方法将大鼠肝癌细胞CBRH7919分为2组:A组用80μmol·L~(-1)的白藜芦醇处理,B组加入等量的二甲基亚砜(DMSO);选取大鼠正常肝细胞BRL-3A作为对照组,也将细胞分为2组:C组用80μmol·L~(-1)的白藜芦醇处理,D组加入等量的DMSO;比较各组细胞的细胞活性、细胞周期变化、caspase-3、bcl-2和bax的表达水平和线粒体膜电位。结果 A组细胞的细胞活力较B组细胞的活力明显减弱(P<0.05)。A、B、C、D组细胞G0/G1分别为(35.67±3.38)%,(48.12±5.01)%,(41.98±4.05)%,(41.87±4.11)%。A组细胞处于G_0/G_1期细胞比例明显少于B组(P<0.05),且2组细胞的caspase-3、bcl-2和bax的表达水平和线粒体膜电位有明显差异(P<0.05)。C组和D组细胞的细胞活性、细胞周期变化、caspase-3、bcl-2和bax的表达水平和线粒体膜电位无明显差异(P>0.05)。结论白藜芦醇可有效诱导大鼠肝癌细胞CBRH7919的凋亡,其可能是通过调控Bcl-2/Bax的表达和引起线粒体膜的去极化发挥抗肿瘤作用。
Objective To study the effect of resveratrol on the apoptosis of hepatoma cell line CBRH7919 in rats. Methods Rat liver cancer cells CBRH7919 were divided into two groups: A group treated with 80μmol·L -1 resveratrol, B group added the same amount of dimethyl sulfoxide (DMSO); selected rats normal liver Cells BRL-3A as a control group, cells were also divided into two groups: C treated with 80μmol·L -1 resveratrol, D group added the same amount of DMSO; compared the cell activity of each group, Cell cycle changes, caspase-3, bcl-2 and bax expression levels and mitochondrial membrane potential. Results The cell viability of group A was significantly weaker than that of group B (P <0.05). The G0 / G1 of group A, B, C and D were (35.67 ± 3.38)%, (48.12 ± 5.01)%, (41.98 ± 4.05)% and (41.87 ± 4.11)%, respectively. The percentage of cells in group G at G 0 / G 1 phase was significantly lower than that in group B (P <0.05), and the expression levels of caspase-3, bcl-2 and bax were significantly different from that of mitochondrial membrane potential (P <0.05) . The cell viability, cell cycle, the expression of caspase-3, bcl-2 and bax and the mitochondrial membrane potential in group C and group D were not significantly different (P> 0.05). Conclusion Resveratrol can effectively induce the apoptosis of rat hepatoma cell line CBRH7919, which may play an antitumor effect through regulating the expression of Bcl-2 / Bax and depolarizing the mitochondrial membrane.
