目的:建立柴芩清肝方提取物中柴胡皂苷a的含量测定方法。方法:采用Diamonsil C18柱(4.6 mm×200 mm,5μm),以乙腈-水为流动相进行梯度洗脱,流速1.0 mL·min-1,蒸发光散射检测器进行检测,漂移管温度105℃,氮气流速2.0L·min-1。结果:柴胡皂苷a进样量在1.7~8.5μg与峰面积对数线性关系良好(r=0.999 4),平均回收率为96.6%,RSD1.8%(n=5)。结论:方法专属性强、准确、快速,适用于柴芩清肝方提取物中柴胡皂苷a的含量测定。
Objective: To establish a method for the determination of saikosaponin a in Chaiqin Qinggan decoction. METHODS: Diamonsil C18 column (4.6 mm × 200 mm, 5 μm) was used for gradient elution with acetonitrile-water as mobile phase at a flow rate of 1.0 mL · min-1. Evaporative light scattering detector The nitrogen flow rate was 2.0 L · min-1. Results: The logarithm of saikosaponin a was 1.7 ~ 8.5μg and the logarithm of peak area was good (r = 0.999 4). The average recovery was 96.6% and the RSD was 1.8% (n = 5). Conclusion: The method is specific, accurate and rapid and suitable for the determination of saikosaponin a in Chaiqin Qinggan Decoction.
