目的研究烟曲霉菌菌丝片段和孢子对气道上皮细胞的作用。方法烟曲霉菌菌丝片段和照射灭活的孢子分别与气道上皮细胞系A549共培养,酶联免疫吸附实验(ELISA)检测A549细胞释放到培养液内的白细胞介素(IL)-6和IL-8。结果菌丝片段刺激A549细胞产生有限的IL-6、IL-8;孢子抑制A549释放IL-6。结论烟曲霉菌菌丝片段和孢子被吸入气道后,致少量细胞因子甚至抑制细胞因子产生,不能使足够的炎症细胞向局部趋化,逃避吞噬,长期存活、芽生、释放大量过敏原。
Objective To study the effects of Aspergillus fumigatus mycelium fragments and spores on airway epithelial cells. Methods The mycelium of Aspergillus fumigatus and the spores inactivated by irradiation were co-cultured with airway epithelial cell line A549. The levels of interleukin (IL) -6 released from A549 cells into culture medium were detected by enzyme-linked immunosorbent assay (ELISA) IL-8. Results mycelium stimulation of A549 cells produce limited IL-6, IL-8; spores inhibit A549 release of IL-6. Conclusion Aspergillus fumigatus mycelial fragments and spores were inhaled into the airways, resulting in a small amount of cytokines and even cytokines, not enough inflammatory cells to local chemotaxis, escape phagocytosis, long-term survival, budding, releasing a large number of allergens.
