激光扫描共焦显微镜(LSCM)可直接、无创性地对组织进行连续光学切片,观察同一标本表面和内部结构。为探索LSCM在内耳形态学研究领域的应用前景,应用LSCM结合免疫荧光组织化学双标记技术,观察5只豚鼠耳蜗毛细胞及支持细胞上部构像。通过毒伞素标记毛细胞肌动蛋白纤维,以抗角质蛋白单克隆抗体标记支持细胞。在LSCM下可清楚观察到毛细胞静纤毛、表皮板以及毛细胞顶部无表皮板区;可同时观察到外柱细胞头板、Deiter细胞指状突顶板以及位于网状层下方的Deiter细胞指状突。LSCM为研究Corti器形态增加了一种新的手段。
Laser scanning confocal microscopy (LSCM) allows direct and noninvasive sequential optical sectioning of the tissue to visualize the surface and internal structure of the same specimen. To explore the potential applications of LSCM in morphological studies of the inner ear, LSCM and immunofluorescence histochemistry double labeling were used to observe the cochlear hair cells and the supracharacteristic of the supporting cells in five guinea pigs. Marked by hairpin actin fibers by phalloidin, supporting cells were labeled with an anti-keratin monoclonal antibody. Under the LSCM, the positive cells of ciliated cells, epidermis and the top of hair cells were clearly observed. The outer plate of Deuter cell, Deiter cell finger top plate and Deiter cells located below the reticular layer were also observed sudden. LSCM for the study of Corti morphology added a new means.
