目的观察丹皮酚对大鼠局灶性脑缺血再灌注损伤后Toll样受体4(TLR4)和神经细胞凋亡的影响,并探讨其作用机制。方法大鼠随机分为假手术组、脑缺血再灌注模型组、尼莫地平组(0.4 mg.kg-1)、丹皮酚组(15、30 mg.kg-1)。采用线栓法阻塞大脑中动脉建立大鼠局灶性脑缺血再灌注模型。通过HE染色观察大鼠大脑皮层区细胞损伤程度,免疫组化法检测缺血侧脑组织TLR4表达的变化,酶联免疫吸附法测定血清白细胞介素6(IL-6)含量,流式细胞仪检测神经细胞凋亡。结果丹皮酚可明显改善大鼠皮层区细胞损伤,减少TLR4的表达、降低血清中IL-6含量,减轻神经细胞凋亡率。结论丹皮酚能减轻缺血再灌注脑组织损伤程度、降低神经细胞凋亡率,机制可能与抑制脑组织内TLR4表达有关。
Objective To observe the effect of paeonol on the expression of Toll-like receptor 4 (TLR4) and neuronal apoptosis after focal cerebral ischemia-reperfusion injury in rats and its mechanism. Methods The rats were randomly divided into sham operation group, model group of cerebral ischemia-reperfusion, nimodipine group (0.4 mg.kg-1) and paeonol group (15, 30 mg.kg-1). A rat model of focal cerebral ischemia / reperfusion was established by occlusion of middle cerebral artery by thread occlusion. The degree of cell damage in the cerebral cortex was observed by HE staining. The expression of TLR4 in the ischemic brain tissue was detected by immunohistochemical method. The content of serum IL-6 was measured by enzyme-linked immunosorbent assay (FACS) Detection of neuronal apoptosis. Results Paeonol could obviously improve the cell injury in cortex of rats, decrease the expression of TLR4, decrease the content of IL-6 in serum and reduce the apoptosis rate of nerve cells. Conclusion Paeonol can reduce the degree of cerebral ischemia-reperfusion injury and reduce the rate of apoptosis of nerve cells, the mechanism may be related to the inhibition of TLR4 expression in brain tissue.
