目的研究“痰瘀同治”方对多囊卵巢综合征模型大鼠胰岛素抵抗的作用。方法采用硫酸普拉睾酮钠注射联合高脂饲料喂养诱导的造模方法制备伴胰岛素抵抗的多囊卵巢综合征大鼠模型,通过光镜计算卵巢排卵情况,测量空腹胰岛素(FINS)和血糖(FBG),并计算胰岛素敏感指数(ISI),采用放射免疫法测定性激素[血清睾酮(T)、雌二醇(E2)、促卵泡刺激素(FSH)和黄体生成素(LH)]的水平来评估“痰瘀同治”方对多囊卵巢综合征模型大鼠胰岛素抵抗的影响。结果与对照组比较,模型组平均排卵数、排卵率及ISI、FSH均显著降低(P<0.01),血清T,E2和LH含量均显著升高(P<0.01),FBG,FINS显著升高,囊性扩张卵泡率显著增加(P<0.05);与模型组比较,治疗组平均排卵数及排卵率显著增加,ISI显著升高,FINS显著降低(P<0.01),血清T和E2含量均显著降低,FSH含量显著升高(P<0.05)。结论 “痰瘀同治”方对伴胰岛素抵抗的多囊卵巢综合征模型大鼠具有改善卵巢排卵障碍、增加胰岛素敏感性和降低性激素水平的作用。
Objective To study the effect of “phlegm and blood stasis with Tongzhi” prescription on insulin resistance in rats with polycystic ovary syndrome. Methods The rat model of polycystic ovary syndrome with insulin resistance was prepared by injection of sodium pramipexate sulfate combined with high-fat diet. Ovulation was measured by light microscope. The levels of fasting insulin (FINS) and blood glucose ), And the insulin sensitivity index (ISI) was calculated and the levels of sex hormones [serum testosterone (T), estradiol (E2), follicle stimulating hormone (FSH) and luteinizing hormone (LH)] were measured by radioimmunoassay Effect of Phlegm and Blood Stasis Tongzhi Decoction on Insulin Resistance in Rats with Polycystic Ovary Syndrome. Results Compared with the control group, the average number of ovulation, the ovulation rate, ISI and FSH of the model group were significantly decreased (P <0.01), the levels of T, E2 and LH were significantly increased (P <0.01) (P <0.05). Compared with the model group, the average ovulation number and ovulation rate of the treatment group increased significantly, the ISI increased significantly, the FINS decreased significantly (P <0.01), and the levels of serum T and E2 (P <0.05), while FSH content increased significantly (P <0.05). Conclusion “Phlegm and blood stasis with Tongzhi” prescription can improve the ovarian ovulation disorder, increase insulin sensitivity and reduce the level of sex hormone in polycystic ovarian syndrome rats with insulin resistance.
