目的:观察加味四逆散及其拆方药物血清对瘦素刺激肝星状细胞增殖的影响。方法:给正常大鼠灌胃加味四逆散及其拆方药物煎剂7天,分别制备各药物含药血清,并将各药物血清用培养基配制成5%、10%、20%、40%4个浓度;培养肝星状细胞,取对数生长期细胞,使用噻唑兰(MTT)比色法检测各药物血清对100ng/mL的瘦素刺激HSC-T6增殖的影响。结果:实验结果显示加味四逆散、疏肝药、活血药、健脾药各药物血清均对增殖的肝星状细胞有抑制作用(P<0.05),在5%~40%的血清浓度范围内,抑制作用呈剂量依赖性关系;秋水仙碱对照组亦能抑制HSC-T6细胞的增殖(P<0.05),但在10%~40%的血清浓度范围内加味四逆散组对HSC-T6增殖的抑制率显著高于秋水仙碱对照组(P<0.05);在10%~40%浓度范围内加味四逆散全方药物血清与3个拆方药物血清相比较,其抑制率明显高于拆方3组,且有显著性差异(P<0.05);3个拆方组中,对增殖细胞的抑制率疏肝药优于活血药优于健脾药,但3组间无显著性差异(P>0.05);3个拆方组与秋水仙碱对照组相比较无统计学意义(P>0.05)。结论:①加味四逆散药物血清及其拆方药物血清均有抑制瘦素刺激HSC-T6增殖的作用;②加味四逆散全方药物血清的抑制作用显著优于拆方中单一方的疗效,说明综合运用疏肝健脾、活血化瘀治疗肝纤维化会取得比单一疏肝或健脾或活血更好的效果。
Objective: To observe the effect of Jiawei Sini San and its disassembled prescription serum on the proliferation of hepatic stellate cells stimulated by leptin. Methods: The normal rats were given gastric gavage and decoction decoction for 7 days to prepare the medicated serum of each drug, and the serum of each drug was formulated into 5%, 10%, 20%, 40% % 4 concentration. Hepatic stellate cells were cultured and logarithmic growth phase cells were harvested. The effect of each serum on the proliferation of HSC-T6 stimulated by leptin of 100 ng / mL was determined by MTT assay. Results: The experimental results showed that all the serums of Jiawei Sini San, Shugan Yao, Huoxueyao and Jianpi Decoction could inhibit the proliferation of hepatic stellate cells (P <0.05). In the serum concentration range of 5% ~ 40% (P <0.05). But colchicine group also inhibited the proliferation of HSC-T6 cells in a dose-dependent manner (P <0.05). However, in the serum concentration range of 10% -40% The inhibitory rate of T6 proliferation was significantly higher than that of colchicine control group (P <0.05). The inhibitory rate of Jiawei Sini San Decoction serum was significantly higher than that of the three disassembled drug serums in the concentration range of 10% ~ 40% (P <0.05). The inhibitory rates of proliferative cells in three groups were better than that of Huoxueyao, but there was no significant difference among the three groups (P> 0.05). There was no significant difference between the three groups (P> 0.05) and the colchicine control group (P> 0.05). Conclusion: ①Jiweisinisan serum and its disassembled serum can inhibit the proliferation of HSC-T6 stimulated by leptin; ② The inhibitory effect of Jiaweisinni Decoction serum is better than that of unilateral prescription , Indicating the comprehensive use of Shuganjianpi, blood circulation to treat liver fibrosis will achieve better than a single Shugan or spleen or blood circulation better results.
