为观察不同浓度紫杉醇诱导犬乳腺肿瘤细胞凋亡的形态学变化,以体外培养犬乳腺肿瘤系CHMm株为模型,分别用0.01、0.1、1μmol/L紫杉醇与犬乳腺肿瘤细胞作用48h后,分别用普通光学显微镜观察细胞形态变化、BrdU标记法载玻片观察细胞增殖状态、AO/EB双荧光染色荧光显微镜及电镜观察犬乳腺肿瘤细胞的凋亡情况。结果显示,不同浓度紫杉醇作用后,CHMm细胞生长受到不同程度的抑制,并随浓度的增加,出现典型的凋亡形态。结果表明,紫杉醇能抑制CHMm生长,引起细胞损伤,诱导细胞凋亡,并具有浓度依赖性。
To observe the morphological changes of apoptosis of canine mammary tumor cells induced by paclitaxel in different concentrations, the in vitro culture of canine mammary gland tumor strain CHMm strain was used as model. After being treated with 0.01,0.1,1μmol / L paclitaxel and canine mammary tumor cells for 48h, The morphological changes of cells were observed under light microscope. The proliferation of cells was observed by BrdU labeling method. The apoptosis of canine mammary tumor cells was observed by AO / EB fluorescent microscope and electron microscope. The results showed that different concentrations of paclitaxel, CHMm cell growth was inhibited to varying degrees, and with the increase in concentration, typical apoptotic morphology. The results showed that paclitaxel can inhibit CHMm growth, cause cell damage, induce apoptosis, and in a concentration-dependent manner.
