目的 探讨异丙肾上腺素(ISP)对内皮素基因表达的影响。方法 通过ISP心肌损伤模型,用放免及分子生物学的方法测定血浆内皮素-1(ET-1)放免活性及心肌和主动脉 ET-1m RNA 的表达。结果 ISP致血浆中ET-1 放免活性明显升高,使心肌及主动脉ET-1 m RNA 高表达。结论 表明ISP使血管内皮细胞ET-1 m RNA 高表达,内皮素分泌增多可能是其致心肌损伤的一个重要原因。
Objective To investigate the effect of isoproterenol (ISP) on endothelin gene expression. Methods The model of myocardial injury was established by ISP, and the radioimmunoassay and molecular biological methods were used to determine the radioactivities of endothelin-1 (ET-1) and the expression of ET-1m RNA in myocardium and aorta. Results The plasma ET-1 radioimmunoassay significantly increased in ISP, leading to high expression of ET-1 mRNA in myocardium and aorta. The results showed that ISP could increase the expression of ET-1 m RNA and increase the secretion of endothelin in vascular endothelial cells, which may be an important cause of myocardial damage.
