目的:观察栀子浸膏对兔膝骨关节炎模型中关节软骨病理改变及关节液、血清中IL-1β表达的影响。方法:24只大白兔随机分为给药组、模型组和对照组(n=8)。给药组、模型组行Hulth法复制骨关节炎模型,术后第5周,给药组以栀子浸膏600mg/kg灌胃,模型组与对照组给予等量生理盐水灌胃,给药3周后处死动物,分别对软骨肉眼下行Pelletier评分及光镜下行Mankin评分,取股骨内侧髁关节软骨组织块行电镜、H-E染色观察软骨细胞形态及检测关节液与血清中IL-1β的含量。结果:栀子浸膏能延缓骨关节炎关节软骨病理损伤,给药组关节液与血清中IL-1β的含量明显低于模型组(P<0.05或P<0.01)。结论:栀子浸膏可能通过降低骨关节炎模型关节液与血清炎症因子IL-1β的表达发挥保护关节软骨的作用,从而延缓骨关节炎病程的进展。
OBJECTIVE: To observe the effect of Gardenia extract on the pathological changes of articular cartilage and the expression of IL-1β in synovial fluid and serum in the rabbit knee osteoarthritis model. Methods: Twenty-four white rabbits were randomly divided into drug administration group, model group and control group (n=8). Hulth method was used to replicate osteoarthritis model in the drug-administered group and the model group. At the 5th week after the operation, the mice in the drug-administered group were administrated with 600mg/kg of wolfberry extract, and the model group and the control group were given intragastric administration with the same volume of saline. Three weeks later, the animals were sacrificed. The Pelletier score and the Mankin score were measured by light microscopy. The chondrocytes of the medial femoral condylar cartilage were observed by electron microscopy and HE staining to observe the morphology of chondrocytes and the levels of IL-1β in the synovial fluid and serum. RESULTS: Gardenia extract could delay the pathological damage of osteoarthritic articular cartilage. The content of IL-1β in synovial fluid and serum of the treatment group was significantly lower than that of the model group (P<0.05 or P<0.01). Conclusion: Gardenia extract may play a role in protecting articular cartilage by reducing the expression of synovial fluid and serum IL-1β in the osteoarthritis model, thereby delaying the progression of osteoarthritis.
