目的 研究氨基葡萄糖及其盐酸盐对白血病细胞K5 6 2的诱导分化效果。方法 利用hexamethylenebisac etamide (HMBA)、氨基葡萄糖及其盐酸盐 ,分别用不同浓度对白血病K5 6 2细胞进行诱导分化实验 ,通过联苯胺、萘酚AS D氯醋酸酯酶及吉姆萨 瑞氏染色、电镜观察和流式细胞仪证实其分化方向。结果 氨基葡萄糖及其盐酸盐对K5 6 2细胞在 0 5mmol·L-1浓度作用 2d后有良好的诱导分化效果 ,诱导K5 6 2向巨噬细胞分化并能使K5 6 2细胞周期阻滞于G0 /G1期 ,与HMBA相比 ,氨基葡萄糖和盐酸盐在作用浓度和作用时间上都有明显优势。结论 氨基葡萄糖及其盐酸盐都可能成为新的高效、低毒的治疗髓系白血病的候选药物
Objective To study the effect of glucosamine and its hydrochloride on the differentiation of leukemia cells K562. Methods Hexamethylenebisac etamide (HMBA), glucosamine and its hydrochloride were used to induce differentiation of leukemia K562 cells by different concentrations. Benzidine, Naphthol AS D-chloroacetate esterase and Jamsar stain, Electron microscopy and flow cytometry confirmed the direction of their differentiation. Results Glucosamine and its hydrochloride had a good effect on differentiation of K562 cells exposed to 0-5mmol·L-1 for 2 days, inducing K562 to differentiate into macrophages and arresting K562 cell cycle In G0 / G1 phase, compared with HMBA, glucosamine and hydrochloride in the concentration and duration of action have obvious advantages. Conclusion Glucosamine and its hydrochloride may be the new drug candidates for myeloid leukemia
