目的:观察IL-2 基因修饰对小鼠腹腔巨噬细胞免疫效应功能的影响。方法:通过重组腺病毒介导,在体外将IL-2基因转染至小鼠腹腔巨噬细胞,检测IL-2 基因修饰的巨噬细胞TNF,IL-1 和NO分泌水平及其杀伤活性。结果:IL-2基因修饰的巨噬细胞TNF,IL-1 和NO分泌水平显著提高,对巨噬细胞敏感株L1210 细胞以及Renca 细胞的杀伤活性显著增强。结论:IL-2基因修饰可增强巨噬细胞的细胞毒效应。本研究为IL-2 基因修饰的巨噬细胞治疗肿瘤提供了实验依据。
Objective: To observe the effect of IL-2 gene modification on the immune effect of mouse peritoneal macrophages. METHODS: IL-2 gene was transfected into peritoneal macrophages of mice by recombinant adenovirus-mediated transfection. IL-2 gene-modified macrophages were examined for the secretion of TNF, IL-1, and NO and their killing activity. RESULTS: Secretion levels of TNF, IL-1 and NO in macrophages modified by IL-2 gene were significantly increased, and the killing activity of macrophage-sensitive strains L1210 cells and Renca cells was significantly enhanced. Conclusion: IL-2 gene modification can enhance the cytotoxic effect of macrophages. This study provided experimental basis for IL-2 gene-modified macrophages to treat tumors.
