目的:通过胶原型诱导型关节炎(CIA)大鼠模型,初步研究维药野西瓜对类风湿性关节炎的抗炎机制。方法:构建II型胶原诱导型关节炎大鼠模型,观察维药野西瓜治疗前后CIA大鼠足踝肿胀程度及踝关节组织病理的改变;ELISA法检测大鼠血清中TNF-α表达水平的变化;免疫组化法检测大鼠足踝关节组织中TNF-α的表达。结果:维药野西瓜可有效缓解CIA大鼠关节的肿胀情况,减缓组织炎症发展;药物作用25d后,与模型组血清中TNF-α含量(60.8±13.5)ng/m L相比,维药野西瓜处理组血清中TNF-α的含量(43.9±2.6)ng/m L明显降低;与模型组相比,维药野西瓜治疗组大鼠足踝关节组织中TNF-α的表达也明显减少。结论:维药野西瓜对CIA大鼠具有较好的抗RA作用,其作用机制可能与降低血清及组织中的TNF-α水平有关。
OBJECTIVE: To study the anti-inflammatory mechanism of wild watermelon on rheumatoid arthritis by collagen induced arthritis (CIA) rat model. Methods: The type II collagen-induced arthritis rat model was established. The degree of ankle swelling and ankle joint pathological changes in CIA rats were observed before and after treatment. The changes of TNF-α in serum were detected by ELISA Immunohistochemistry was used to detect the expression of TNF-α in the foot and ankle joint tissues of rats. Results: After the drug effect for 25 days, compared with the level of TNF-α in serum of model group (60.8 ± 13.5) ng / m L, the drug of Weiye watermelon could effectively relieve the swelling of joint in CIA rats and relieve the development of tissue inflammation. The serum level of TNF-αin wild watermelon treated group was significantly lower than that of the model group (43.9 ± 2.6) ng / m L; the expression of TNF-α in the foot and ankle joint of the treated group was also significantly decreased . Conclusion: Weiye watermelon has a good anti-RA effect on CIA rats, and its mechanism may be related to the reduction of serum and tissue levels of TNF-α.
