应用昆虫杆状病毒表达系统 (BES)在昆虫细胞 Tn- 5 B1- 4中高效表达了人胶质细胞源性神经营养因子 (GDNF) ,PAGE分析表达量占细胞可溶性蛋白质的 30 %左右 ,表达产物经亲和层析纯化后纯度达 80 %以上。活性研究表明 ,昆虫细胞表达的 GDNF蛋白能显著促进多巴胺能神经元的存活。此研究为进一步研究 GDNF结构与功能打下了良好的基础。
The human glial cell line-derived neurotrophic factor (GDNF) was highly expressed in the insect cell line Tn-5 B1-4 using the insect baculovirus expression system (BES). The expression level of GDNF was about 30% of that of soluble protein Purification of the product by affinity chromatography purity of more than 80%. Activity studies show that GDNF protein expressed by insect cells can significantly promote the survival of dopaminergic neurons. This study laid a good foundation for further study on the structure and function of GDNF.
