目的探讨超声联合紫杉醇微泡剂对人卵巢癌细胞COC1的增殖抑制作用。方法将人卵巢癌细胞株COC1随机分为6组:对照组、空白微泡剂+超声组、紫杉醇微泡剂组、单纯紫杉醇组、紫杉醇+超声组、紫杉醇微泡剂+超声组。观察各组处理后癌细胞的增殖、细胞形态及细胞凋亡情况。结果紫杉醇微泡剂+超声组COC1细胞增殖被明显抑制,24,48,72 h的抑制率分别为(43.60±2.64)%,(37.89±1.30)%,(28.95±2.89)%。电镜观察结果显示,紫杉醇微泡剂+超声组的COC1细胞数量减少最显著(P<0.05),细胞折光度差,细胞固缩,出现许多细胞碎屑,PI/hoechst染色检测显示细胞凋亡率最高,为(52.67±9.29)%(P<0.05)。结论超声能有效增强紫杉醇微泡剂对人卵巢癌细胞COC1的杀伤能力。
Objective To investigate the inhibitory effect of ultrasound combined with paclitaxel on the proliferation of human ovarian cancer cells COC1. Methods Human ovarian cancer cell line COC1 was randomly divided into 6 groups: control group, blank microbubbles + ultrasound group, paclitaxel microbubbles group, paclitaxel group, paclitaxel + ultrasound group, paclitaxel microbubbles + ultrasound group. The proliferation, cell morphology and apoptosis of cancer cells in each group were observed. Results The proliferation of COC1 cells was significantly inhibited in paclitaxel microbubbles + ultrasound group. The inhibition rates of COC1 cells were (43.60 ± 2.64)%, (37.89 ± 1.30)% and (28.95 ± 2.89)% at 24, 48, and 72 h, respectively. Electron microscopy results showed that the number of COC1 cells in paclitaxel microbubbles + ultrasound group decreased most significantly (P <0.05), cell refractive index was poor, cell shrinkage was observed, and many cell debris appeared. PI / hoechst staining showed that the apoptosis rate The highest was (52.67 ± 9.29)% (P <0.05). Conclusion Ultrasound can effectively enhance paclitaxel microbubbles on human ovarian cancer cells COC1 killing capacity.
