通常,一根色谱柱在分析数千个样品之后性能仍然保持良好,但也有的柱仅分析不多的样品后几乎就报废了。影响柱寿命和其它问题的因素很多,而有些因素是操作者很难控制的,如果被分析的样品(如分析生物样品),怎么净化样品也是“脏”的,好于色谱柱的影响是非常大的。然而采取下列措施后,在多数情况下总能够人为地减少柱上故障,达到延长柱寿命的目的。
Typically, a column still performs well after analyzing thousands of samples, but some columns are almost scrapped after only a small sample has been analyzed. There are a number of factors that affect column lifetime and other issues, some of which are difficult to control by the operator. If the sample being analyzed (eg, analyzing a biological sample), it is also “dirty” to clean the sample, which is better than the column big. However, following the following measures, in most cases can always reduce the man-made fault on the column, to extend the life of the purpose of the column.
