目的 检验人参皂甙Rb1,Rg1和维生素E是否能抑制次黄嘌呤 (HX) /黄嘌呤氧化酶 (XO)对培养的牛视网膜色素上皮 (RPE)细胞的氧化损伤作用。方法 建立牛眼RPE细胞培养体系以及RPE细胞氧化损伤模型。利用MTT法观察人参皂甙Rb1、Rg1和维生素E对氧化损伤的牛RPE细胞增殖的影响。结果 加入不同浓度的Rb1后测得的细胞OD值普遍较HX/XO组的OD值高 ,但无显著性差异 ;当Rg1=0 1和 1μg/ml,维生素E =1和 10 μg/ml时测得的OD值与HX/XO组的OD值相比有显著性差异 (P <0 0 5 )。结论 Rg1和维生素E能对抗氧自由基对体外培养的牛RPE细胞生长的抑制作用 ,而Rb1的作用不明显。
Objective To investigate whether ginsenoside Rb1, Rg1 and vitamin E could inhibit the oxidative damage of cultured bovine retinal pigment epithelium (RPE) cells induced by hypoxanthine (HX) / xanthine oxidase (XO). Methods Bovine RPE cell culture system and RPE cell oxidative damage model were established. The effects of ginsenoside Rb1, Rg1 and vitamin E on the proliferation of oxidatively injured bovine RPE cells were observed by MTT assay. Results The cell OD values measured after addition of different concentrations of Rb1 were generally higher than those of HX / XO OD groups, but no significant difference was found. When Rg1 = 0 1 and 1 μg / ml, vitamin E = 1 and 10 μg / ml The measured OD value was significantly different from that of HX / XO group (P <0 05). Conclusion Rg1 and vitamin E can antagonize the inhibitory effect of oxygen free radicals on the growth of bovine RPE cells cultured in vitro, while the effect of Rb1 is not obvious.
