肿瘤转移是恶性肿瘤最为重要的生物学特征之一,同时也是影响预后及导致治疗失败的主要因素。近年来研究表明,肿瘤微环境中的胞外基质(extracellular matrix,ECM)纤维生成在肿瘤转移过程中发挥极为关键的作用。肿瘤ECM纤维生成会形成相互交联的网状结构,这种网状结构不仅对肿瘤细胞起到支持和营养作用,同时对肿瘤的生长和浸润也起到相当重要的作用。因此,阻断肿瘤组织ECM纤维生成有可能对肿瘤转移产生抑制作用,这使得ECM纤维生成作为治疗靶标,备受人们关注。该文对肿瘤ECM纤维生成与肿瘤转移研究进展进行综述,并对靶向肿瘤ECM纤维生成进行肿瘤转移治疗的策略进行探讨,为后续研究及指导临床提供重要参考。
Tumor metastasis is one of the most important biological characteristics of malignant tumors, but also affect the prognosis and lead to treatment failure of the main factors. In recent years, studies have shown that the extracellular matrix (ECM) fiber in the tumor microenvironment plays an extremely crucial role in tumor metastasis. Tumor ECM fiber formation will form a cross-linked network structure, this network structure not only play a supporting and nutritional role of tumor cells, but also on tumor growth and infiltration also played a very important role. Therefore, blocking tumor tissue ECM fiber production may have an inhibitory effect on tumor metastasis, which makes ECM fiber production as a therapeutic target for much attention. This article reviews the progress of tumor ECM fibrogenesis and tumor metastasis, and discusses the strategy of tumor metastasis targeting tumor ECM fibrogenesis, which will provide important reference for further research and clinical guidance.
