实验用 FOS免疫组化方法 (ABC法 )研究了缺血诱导的大鼠视网膜内 c- fos原癌基因的表达情况。实验动物用升高眼压的方法做成视网膜缺血模型 ,依缺血后存活时间不同分 1 5′、 30′、 1 h、 2 h、 4h、 6 h、 1 2 h七个组 ,每只大鼠右眼为缺血眼 ,左眼做自身对照眼 ,另设正常对照组。动物腹腔麻醉 ,4%多聚甲醛灌注固定 ,取双眼做冰冻切片 ,片厚 1 5μm。实验结果显示缺血后 1 5′组大鼠视网膜内核层最先出现少量卵圆型浅棕色的 FOS阳性神经元胞核 ,30分钟至 1 h FOS表达逐渐增强 ,节细胞层也出现 FOS阳性胞核。缺血后 2 h FOS表达达最高峰 ,缺血后 4h FOS阳性胞核逐渐减少 ,1 2 h达正常组水平。自身对照眼及正常对照眼视网膜节细胞层偶见 FOS阳性胞核
The expression of c-fos proto-oncogene in rat’s retina induced by ischemia was studied by FOS immunohistochemistry (ABC method). Experimental animals with elevated intraocular pressure retinal ischemia model made according to the survival time after ischemia according to different points 1 5 ’, 30’, 1 h, 2 h, 4 h, 6 h, 12 h seven groups, each Only right eye for ischemic eyes, the left eye to do their own control eye, the other set up a normal control group. The experimental results showed that a small amount of oval light brown FOS positive neurons appeared first in the rat retina after 15min ischemia. The expression of FOS positive neurons gradually increased from 30min to 1h, and FOS positive cells appeared in the ganglion cells nuclear. The expression of FOS peaked at 2 h after ischemia, and the number of FOS positive nuclei gradually decreased 4 h after ischemia, reaching the level of normal group at 12 h.
