目的研究结直肠癌中Wnt/β-联蛋白(catenin)信号通路调控因子分泌型卷曲相关蛋白(SFRP)2基因启动子区甲基化状态及β-catenin蛋白的表达模式,探讨两者在结直肠癌中的关系及意义。方法用甲基化特异性聚合酶链反应(MSP)法和免疫组织化学法分别检测结直肠癌组、腺瘤组、非腺瘤性息肉组、健康对照组中SFRP2基因启动子区甲基化状态和β-catenin蛋白的表达情况。结果在结直肠癌组、腺瘤组、非腺瘤性息肉组、健康对照组中,SFRP2基因启动子区甲基化率分别为79%、63%、8%、0,差异有统计学意义(P<0.05);β-catenin蛋白表达率分别为74%、48%、4%、0,差异有统计学意义(P<0.05);SFRP2启动子区甲基化与β-catenin异常表达之间存在相关性(P<0.01);SFRP2启动子区甲基化和β-catenin异常表达均与肿瘤分化程度有关(P<0.05)。结论 SFRP2启动子区域甲基化频率及β-catenin异常表达率在结直肠癌中上调,两者可能共同促进了结直肠癌的发生发展。
Objective To investigate the methylation status of β-catenin protein and the promoter region of Wnt / β-catenin signaling pathway regulator (SFRP) 2 gene in colorectal cancer, Relationship and Significance of Rectal Cancer. Methods Methylation-specific polymerase chain reaction (MSP) and immunohistochemistry were used to detect the promoter methylation of SFRP2 gene in colorectal cancer, adenoma, non-adenomatous polyps and healthy controls Status and β-catenin protein expression. Results The methylation rates of SFRP2 gene promoter region in colorectal cancer group, adenoma group, non-adenomatous polyp group and healthy control group were 79%, 63%, 8%, 0 respectively, the difference was statistically significant (P <0.05). The expression of β-catenin protein was 74%, 48%, 4%, 0 respectively (P <0.05). The methylation of SFRP2 promoter and the abnormal expression of β-catenin (P <0.01). The methylation status of SFRP2 promoter and the abnormal expression of β-catenin were correlated with tumor differentiation (P <0.05). Conclusions The frequency of methylation and the abnormal expression of β-catenin in SFRP2 promoter region are up-regulated in colorectal cancer, both of which may contribute to the development of colorectal cancer.
