在生化实验中可以利用细胞的比重、体积、表面电荷及对某种材料的粘附性等性质的差异达到分离细胞的目的。常用的方法有蔗糖密度梯度法、Ficoll法、Percoll法等。有人报道用Percoll方法可分离死细胞与活细胞。我们对此进行了试验。1.试剂(1) 瑞典产Percoll。(2) 5mM Tris-HCl~5mM CaCl_2。(3) 1.25M蔗糖Tris缓冲液:取21.3g蔗糖用(2)液溶解至50ml。1MTris调pH至7.2~7.4。(4) 0.25M蔗糖缓冲液:取1.25M蔗糖液用(2)液稀释5倍即成。1MTris调pH至7.2~7.4。

Common methods are sucrose density gradient method, Ficoll method, Percoll method. It has been reported that the Percoll method can be used to separate dead cells and living cells. We tested it. Reagents (1) Swedish production of Percoll. (2) 5 mM Tris-HCl to 5 mM CaCl 2. (3) 1.25M sucrose Tris buffer: Take 21.3g of sucrose and dissolve to 50ml with (2) solution. 1MTris pH adjusted to 7.2 ~ 7.4. (4) 0.25M sucrose buffer: take 1.25M sucrose solution (2) diluted 5 times Serve. 1MTris pH adjusted to 7.2 ~ 7.4.
