以普通小麦中国春、长穗偃麦草及其7个二体异附加系为材料,用26个10碱基随机引物进行随机扩增,以检测普通小麦与长穗偃麦草间的多态性并建立长穗偃麦草染色体特有的RAPD标记。实验结果表明:26个引物在中国春及长穗偃麦中共扩增出180条带,其中中国春121条,长穗偃麦草94条,二者相同的带只有35条,仅占19.44%,另外80.56%的带在二者间揭示了多态性,从分子水平揭示二者基因组间存在着较高水平的多态性。有三个引物OPE—05、OPF—03和OPF—15分别在两亲本间揭示了多态性,同时这一多态性带也分别出现于双二倍体及1E、3E染色体的附加系中,其片段大小分别为1300bp、700bp、400bp,因而OPE—05_(1300)、OPF—03_(700)和OPF—15_(400)分别是1E和3E染色体的特异RAPD标记,可以用来快速地跟踪鉴定1E和3E染色体并应用于偃麦草属的研究中。
A total of 26 10-bp random primers were used to amplify the common wheat Chinese Spring, Eriosphaeria barbarum and its seven isogenic lines to detect the polymorphism between common wheat and Eriosphae alba. Establishment of RAPD Markers Specific to Chromosomes of Agropyron elongatum. The results showed that there were 180 bands amplified by 26 primers in Chinese spring and long-spike euchromium, of which 121 were Chinese and 94 were long-tailed Echinochloa crus-galli. There were only 35 identical bands with only 19.44% In addition, 80.56% of the bands revealed the polymorphism between the two and revealed a high level of polymorphism between the two genomes at the molecular level. Three primers OPE-05, OPF-03 and OPF-15 respectively revealed the polymorphism between the two parents, and the polymorphic bands also appeared in the diploid and additional lines of chromosomes 1E and 3E, respectively. Therefore, OPE-05_ (1300), OPF-03_ (700) and OPF-15_ (400) are specific RAPD markers on chromosomes 1E and 3E, respectively, which can be used for rapid follow-up identification 1E and 3E chromosomes and are used in the research of Agropytis.
