目的 研究10例乳腺癌细胞系的6号染色体长臂缺失。方法 运用荧光原位杂交(fluorescenceinsituhybridization,FISH)技术,并采用37个分布于6q126q27的YAC探针和6号染色体着丝粒特异性探针,检测了10例乳腺癌细胞系的6号染色体长臂。结果 在5例细胞系中发现6q126q27的大段缺失,1例发现有6q25q27的小段缺失。在含有多个细胞亚群的两例细胞系中,6q的缺失仅见于6号染色体多倍体的细胞。在1例细胞系中检测到6q21q22.2部位有DNA扩增。结论 6q25q27是乳腺癌的常见缺失区。6q的缺失可能是继发性改变。
Objective To study the deletion of the long arm of chromosome 6 in 10 breast cancer cell lines. Methods Fluorescence in situ hybridization (FISH) technique was used and 37 YAC probes distributed on 6q126q27 and chromosome 6 centromere specific probes were used to detect chromosome 6 long arms of 10 breast cancer cell lines. . Results The large deletion of 6q126q27 was found in 5 cell lines, and the small deletion of 6q25q27 was found in 1 case. In two cell lines containing multiple cell subpopulations, the 6q deletion was found only in cells of chromosome 6 polyploidy. DNA amplification of the 6q21q22.2 site was detected in 1 cell line. Conclusion 6q25q27 is a common missing region in breast cancer. The absence of 6q may be a secondary change.
