目的建立高效液相色谱法研究大鼠口服西河柳总黄酮后柽柳素和山奈甲黄素的组织分布。方法采用Waters C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-水(pH 2.7)为流动相,流速1.0 mL.min 1,检测波长254 nm,柱温35℃,组织样品用固相萃取法处理。结果在多种组织样品中同时检测到柽柳素和山奈甲黄素,柽柳素在肺、心中分布最高,山奈甲黄素最高分布在心,次之为肺。结论利用高效液相色谱法可以同时进行柽柳素和山奈甲黄素组织分布研究。
OBJECTIVE To establish a high performance liquid chromatography (HPLC) method for the tissue distribution of ralcinose and kaempferol in rats after oral administration of X. Methods A Waters C18 column (4.6 mm × 250 mm, 5 μm) was used with methanol-water (pH 2.7) as the mobile phase at a flow rate of 1.0 mL · min 1. The detection wavelength was 254 nm and the column temperature was 35 ℃. Extractive processing. Results In the samples of various tissues, both ictalis and kaempferol were detected simultaneously. The distribution of ictalisin in lung and heart was the highest, and the highest concentration of kaempferol was in the heart, followed by the lung. Conclusion The study of tissue distribution of ralizumab and kaempferide can be carried out simultaneously by high performance liquid chromatography.
