目的 探讨胶质瘤细胞血小板源生长因子 (PDGF)BB自分泌环活性与肿瘤细胞egr 1基因表达的关系 ,以及它们在胶质瘤发生、发展中的作用。方法 用mRNA原位杂交和免疫组化染色观察了 73例人脑胶质瘤组织。结果 胶质瘤细胞PDGFBmRNA表达水平随肿瘤恶性程度增加而升高。egr 1mRNA、EGR 1蛋白、PDGFα受体和β受体及酪氨酸磷酸化蛋白阳性肿瘤细胞密度彼此间均呈显著性正相关 ,并均随肿瘤恶性程度及肿瘤细胞PDGFBmRNA表达水平升高而同步递增。结论 胶质瘤细胞普遍存在PDGFBB自分泌环活性和egr 1基因表达异常增加 ,它们可能互为因果 ,并产生一个放大效应 ,这也许是胶质瘤发生、发展的重要因素。
Objective To investigate the relationship between autocrine loop activity of platelet derived growth factor (PDGF) BB and the expression of egr 1 gene in glioma cells and their roles in the development and progression of glioma. Methods Totally 73 human glioma tissues were observed by mRNA in situ hybridization and immunohistochemistry. Results The expression of PDGFB mRNA in glioma cells increased with the malignant degree of tumor. The expressions of egr 1 mRNA, EGR 1 protein, PDGFα receptor, β receptor and tyrosine phosphorylation protein positive tumor cells were significantly and positively correlated with each other, and they were all correlated with the degree of tumor malignancy and the increase of PDGFB mRNA expression in tumor cells Increment. Conclusions Glutathione esterase (PDGFBB) has an autocrine loop activity and an abnormal increase of egr 1 gene expression in glioma cells. They may play causative roles in each other and produce a magnifying effect, which may be an important factor in the development and progression of glioma.
