新疆野生冰草抗旱功能基因BADH的克隆及表达载体的构建

【目的】克隆新疆野生冰草抗旱相关基因甜菜碱醛脱氢酶(betaine aldehyde dehydrogenase,BADH)基因,为进一步研究其抗旱功能奠定基础。【方法】利用同源克隆法和RACE方法扩增获得新疆草原区强抗旱植物沙生冰草抗旱相关基因BADH,并将其构建到植物表达载体pBI121上。【结果】克隆获得的新疆野生沙生冰草抗旱相关基因BADH全长1422 bp,编码区序列全长为1182 bp,GenBank注册号为GU181396.1,并成功构建了植物过表达载体。 【Objective】 To clone the gene of drought-related gene betaine aldehyde dehydrogenase (BADH) in Xinjiang, and lay a foundation for further study on drought resistance. 【Method】 The drought resistance-associated gene BADH of Sabina przewalskii, a strongly drought-tolerant plant in Xinjiang grassland, was amplified by homologous cloning and RACE methods and was cloned into plant expression vector pBI121. 【Result】 The results showed that the full-length BADH gene was 1422 bp in length and the coding sequence was 1182 bp in length. The GenBank accession number was GU181396.1. The plant overexpression vector was successfully constructed.
