目的:探讨慢性心力衰竭大鼠β3肾上腺素能受体(β3受体)表达以及不同剂量的卡维地洛对其的干预效果。方法:32只雄性Wistar大鼠分为假手术组(n=10)和心力衰竭组(n=22)。心力衰竭组大鼠分别予生理盐水(心力衰竭对照组,n=6)、低剂量卡维地洛(3mg·kg-1.d-1,n=8)或高剂量卡维地洛(6mg·kg-1.d-1,n=8)口服灌注,共3个月。结果:与假手术组比较,心力衰竭组大鼠左室收缩末压、左室内压最大收缩和舒张速率的绝对值显著降低(P<0.01),左室舒张末压显著增高(P<0.01)。与低剂量卡维地洛组比较,高剂量卡维地洛组LVESP及dp/dtmax明显升高、LVEDP明显降低(P<0.01)。高剂量卡维地洛组的左心室质量指数明显低于低剂量卡维地洛组和心力衰竭对照组(P<0.01)。研究组左心室β3受体表达明显高于假手术组(P<0.01)。高剂量卡维地洛组大鼠左心室β3受体表达明显低于假手术组和低剂量卡维地洛组(P<0.01)。结论:大鼠衰竭左心室的β3受体表达明显增加。高剂量卡维地洛对大鼠心力衰竭后血流动力学、左心室重构的改善作用明显优于低剂量时。高剂量卡维地洛明显抑制大鼠衰竭左心室β3受体表达,而低剂量卡维地洛没有此作用。
Objective: To investigate the expression of β3 adrenergic receptor (β3) in rats with chronic heart failure and the effects of different doses of carvedilol on it. Methods: Thirty-two male Wistar rats were divided into sham operation group (n = 10) and heart failure group (n = 22). Rats in heart failure group were treated with saline (heart failure control group, n = 6), low dose carvedilol (3 mg · kg-1.d-1, n = 8) or high dose carvedilol · Kg-1.d-1, n = 8) orally for 3 months. Results: The absolute value of left ventricular end systolic pressure, maximal systolic and diastolic velocity of left ventricular pressure was significantly decreased (P <0.01) and left ventricular end diastolic pressure was significantly increased in heart failure group compared with sham operation group (P <0.01) . Compared with the low-dose carvedilol group, the LVESP and dp / dtmax in the high-dose carvedilol group were significantly increased and the LVEDP level was significantly decreased (P <0.01). The LV mass index of high-dose carvedilol group was significantly lower than that of low-dose carvedilol group and heart failure control group (P <0.01). Study group left ventricular β3 receptor expression was significantly higher than the sham operation group (P <0.01). The left ventricular β3 receptor expression in high-dose carvedilol group was significantly lower than that in sham-operated group and low-dose carvedilol group (P <0.01). Conclusion: The expression of β3 receptor in left ventricle of rats is obviously increased. The effect of high dose carvedilol on hemodynamics and left ventricular remodeling after heart failure in rats was obviously better than that at low dose. High-dose carvedilol significantly inhibited left ventricular β3 receptor expression in rats, whereas low-dose carvedilol did not.
