目的观察罗勒多糖对乳腺癌细胞系MDA-MB-231侵袭性的影响。方法将体外培养的MDA-MB-231细胞分为实验组和对照组。实验组细胞用不同浓度罗勒多糖(100、300μg/mL)处理,对照组不加任何刺激因子。Transw ell小室检测细胞侵袭能力,RT-PCR、Western blotting方法检测与侵袭相关的基质金属蛋白酶2(MMP-2)、基质金属蛋白酶9(MMP-9)和膜型基质金属蛋白酶1(MT1-MMP)的表达。结果①罗勒多糖可抑制MDA-MB-231细胞的侵袭力(P<0.01);②罗勒多糖抑制MT1-MMP分子的mRNA表达(P<0.05)及下调其蛋白表达水平。③罗勒多糖对MMP-2及MMP-9分子的mRNA表达无影响。结论罗勒多糖可通过下调MT1-MMP的表达抑制MDA-MB-231细胞的侵袭。
Objective To investigate the effect of basil polysaccharide on the invasiveness of breast cancer cell line MDA-MB-231. Methods MDA-MB-231 cells cultured in vitro were divided into experimental group and control group. The experimental group treated with different concentrations of basil (100, 300μg / mL), the control group without any stimulus. Transw ell cells were used to detect cell invasion. RT-PCR and Western blotting were used to detect the expression of MMP-2, MMP-9 and MT1-MMP )expression. Results ① basil could inhibit the invasiveness of MDA-MB-231 cells (P <0.01). ② basil inhibited the mRNA expression of MT1-MMP (P <0.05) and down-regulated the protein expression. ③ basil polysaccharide had no effect on mRNA expression of MMP-2 and MMP-9. Conclusion Basil polysaccharide can inhibit the invasion of MDA-MB-231 cells by down-regulating the expression of MT1-MMP.
