抗癌药喜树碱(camptothecine)系从我国特有植物喜树[Camptotheca acuminata De-cne]中提取的生物碱。国内外药典均未收载,部分省市有它的质量标准,含量测定均采用紫外分光光度法。其原理是喜树碱与碱作用,内酯环破裂生成水溶性钠盐,它的紫外吸收光谱在波长254nm 处有最大吸收。我们在修订湖北省药品标准时,按此法测定原料药含量,重现性不够好,含量往往偏低。为寻其原因,进行了一些试验,发现光和热对含量测定皆有影响。现介绍如下。
The anti-cancer drug camptothecine is an alkaloid extracted from Camptotheca acuminata De-cne. Pharmacopoeia are not included at home and abroad, some provinces and cities have its quality standards, determination of content are used UV spectrophotometry. Its principle is the role of camptothecin and alkali, lactone ring rupture to generate water-soluble sodium salt, its UV absorption spectrum at a maximum wavelength of 254nm absorption. We revised the Hubei Province drug standards, according to this method of determination of API content, reproducibility is not good enough, the content is often low. In order to find out the reason, some experiments were carried out and found that both light and heat have an influence on the content determination. Now introduced as follows.
