我们应用国外新近报道的三种方法,分别从牛血清中提取了K,并对其纯度和活性进行了鉴定,对其产量进行了核算,同时与国外最新的有关报道作了比较。我们的试验结果表明,采用改良的Lachmann方法,即用酵母多糖提取K,再通过DEAE-纤维素层析柱提纯能获得纯度较高(琼脂双扩散试验对全牛血清抗血清出现一条沉淀线)、产量较高的K(1.48mg/100ml牛血清)。层析图谱表明,K的生物学活性存在于最后一峰中。文中K 的活性是采用EAC_(3bi)(牛)的经典胶固反应鉴定的。
We use three newly reported foreign methods, respectively, from the bovine serum extracted K, and its purity and activity were identified, the yield was accounted for, and with the latest foreign reports were compared. Our experimental results show that using improved Lachmann method, that is, extracting K with zymosan and purifying by DEAE-cellulose chromatography can obtain high purity (a precipitation line appears in the whole bovine serum antiserum by the agar double diffusion test) , Yield higher K (1.48 mg / 100 ml bovine serum). The chromatogram shows that the biological activity of K exists in the last peak. The activity of K in this paper is identified by the classical colloidal reaction of EAC_ (3bi) (bovine).
