目的:探讨HOTAIR在胃癌细胞株中的表达情况,探讨竹节香附素A是否可通过下调人胃癌细胞中HOTAIR表达抑制胃癌细胞增殖。方法:采用实时定量PCR检测胃癌细胞株HGC-27、MGC-803、SGC-7901和胃黏膜上皮正常GES-1细胞中HOTAIR的表达。将胃癌MGC-803细胞分别用HOTAIR特异性小干扰RNA、竹节香附素A、siHOTAIR+竹节香附素A处理,并与阴性组对照,分别采用CCK-8法和克隆形成实验检测三组胃癌细胞增殖情况。结果:HOTAIR在胃癌细胞株中高表达,用si-HOTAIR沉默HOTAIR表达后,胃癌细胞增殖得到抑制。竹节香附素A对胃癌细胞增殖呈剂量依赖性。竹节香附素A可下调胃癌细胞MGC-803细胞中HOTAIR表达水平(P<0.05)。竹节香附A与si-HOTAIR联合运用可明显下调HOTAIR表达,与单纯si-HOTAIR处理有统计学差异(P<0.05)。结论:竹节香附素A可能通过下调胃癌MGC-803细胞中HOTAIR的表达从而抑制胃癌细胞增殖。
OBJECTIVE: To investigate the expression of HOTAIR in gastric cancer cell lines and to explore whether sarcandrosin A can inhibit the proliferation of gastric cancer cells by down-regulating HOTAIR expression in human gastric cancer cells. Methods: HOTAIR expression in gastric cancer cell lines HGC-27, MGC-803, SGC-7901 and normal gastric epithelial GES-1 cells was detected by real-time PCR. The gastric cancer MGC-803 cells were treated with HOTAIR-specific small interfering RNA, sarcodictyin A, siHOTAIR + Sarcandra A, respectively, and compared with the negative group, CCK-8 and clonogenic assay were used to detect three groups Proliferation of gastric cancer cells. Results: HOTAIR was highly expressed in gastric cancer cell lines. After HOTAIR was silenced by si-HOTAIR, the proliferation of gastric cancer cells was inhibited. Sarcaxin A has a dose-dependent effect on the proliferation of gastric cancer cells. Scutellain A down-regulated the HOTAIR expression in gastric cancer cell line MGC-803 (P <0.05). The combination of sikfu A and si-HOTAIR could obviously down-regulate the expression of HOTAIR, which was significantly different from si-HOTAIR treatment (P <0.05). CONCLUSION: Sarcandra suppository A may inhibit the proliferation of gastric cancer cells by down-regulating HOTAIR expression in MGC-803 cells.
