目的对芽胞杆菌Bacillus sp.FJAT-46478来源的伊默匹泊(immepip)成分进行分析与检测。方法采用液相-四级杆飞行时间质谱技术(liquid chromatography-hybrid quadrupole time-of-flight mass spectrometry,LC–QTOF-MS)分析芽胞杆菌FJAT-46478发酵液中胞外代谢物的成分。利用Mass Hunter软件,对原始数据进行分子特征提取,通过Metlin代谢物质谱数据库比对获得代谢物的信息。结果筛选获得1株产伊默匹泊(immepip)的芽胞杆菌属菌株Bacillus sp.FJAT-46478,其中伊默匹泊匹配得分为95.6,含量占发酵液总代谢物相对含量的2.44%,保留时间为5.8384 min,精确质量数为165.1262,质量偏差为-4.66×10-5。结论菌株FJAT-46478为伊默匹泊的高产菌株,伊默匹泊是一种重要的组胺H_3受体激动剂,本研究为芽胞杆菌来源伊默匹泊的开发与应用提供实验材料。
Objective To analyze and detect the immepip component of Bacillus sp. FJAT-46478. Methods The constituents of extracellular metabolites in Bacillus sp. FJAT-46478 were analyzed by liquid chromatography-quadrupole time-of-flight mass spectrometry (LC-QTOF-MS). Using Mass Hunter software, the original data were extracted by molecular feature, and the metabolites were obtained through Metlin metabolite database. Results A strain of Bacillus sp. FJAT-46478 producing immepip was screened out, in which the Imeropfen matching score was 95.6, the content accounted for 2.44% of the relative content of total metabolites in fermentation broth, the retention time 5.8384 min, the exact mass number is 165.1262 and the mass deviation is -4.66 × 10-5. Conclusion The strain FJAT-46478 is a high yielding strain of imepirox, and ibuprofen is an important histamine H_3 receptor agonist. In this study, we provided experimental materials for the development and application of ibuprofen.
