以含70%EGCG的茶多酚为原料,单分散15μm的聚苯乙烯-二乙烯基苯微球为分离介质,以安全性较好的乙醇-水为洗脱剂,结合回收率和产率为考察对象,从流动相比例、上样量以及流动相流速3个方面来探索最佳的制备色谱纯化EGCG的工艺条件。在流动相为乙醇/水=30/70(v/v)、上样量为900 mg、流速为10 mL/min的条件下,分离得到EGCG的单体纯度为97.49%,回收率75.81%。该方法具有分离效率高、处理量大、产品安全性好、生产周期短等特点,为高纯度EGCG的工业化生产提供了一种有效的技术支持。
Using 70% EGCG polyphenols as raw material, monodisperse polystyrene-divinylbenzene microspheres with 15μm dispersion as the separation medium and ethanol-water with good safety as eluent, the recovery and the yield As the object of study, the optimum process conditions for the preparation of chromatographic purification EGCG were explored from three aspects: mobile phase ratio, sample loading and mobile phase flow rate. Under the condition of the mobile phase of ethanol / water = 30/70 (v / v), the sample volume of 900 mg and the flow rate of 10 mL / min, the monomer purity of EGCG was 97.49% and the recovery was 75.81%. The method has the characteristics of high separation efficiency, large processing capacity, good product safety and short production cycle, and provides an effective technical support for the industrialized production of high-purity EGCG.
