本研究观察了雌二醇(E2)对培养的人胚肺成纤维细胞增殖的直接和间接调控影响。结果显示:E2可降低肺成纤维细胞的3H-TdR掺入量;E2与肺泡巨噬细胞(AM)培养4h的上清液可使肺成纤维细胞的3H-TdR掺入量显著增加;消炎痛预处理不影响E2的上述作用;E2与AM培养18h可使培养液中内皮素(ET)的含量显著升高,但培养4h对AM的ET分泌的影响不显著。以上结果表明,E2对肺成纤维细胞的增殖具有直接抑制和通过AM而间接促进的双向调控作用,其作用与内源性前列腺素无关。E2间接促进成纤维细胞增殖的机制除与促进AM的ET分泌有关外还有其它细胞因子参与。本结果提示,E2参与肺间质稳态的调控
This study examined the direct and indirect effects of estradiol (E2) on the proliferation of cultured human embryonic lung fibroblasts. The results showed that: E2 can reduce 3H-TdR incorporation of lung fibroblasts; E2 and alveolar macrophages (AM) cultured 4h supernatant of lung fibroblasts 3H-TdR incorporation significantly increased; anti-inflammatory Pain pretreatment did not affect the above effects of E2; E2 and AM cultured 18h can make the content of endothelin (ET) increased significantly, but cultured 4h had no significant effect on ET secretion of AM. The above results show that E2 has a direct inhibition on the proliferation of lung fibroblasts and indirectly through AM to promote bidirectional regulation, its role and endogenous prostaglandin has nothing to do. E2 indirectly promote the proliferation of fibroblasts in addition to promoting the secretion of AM related to the secretion of ET there are other cytokines involved. The results suggest that E2 is involved in the regulation of pulmonary interstitial homeostasis
