目的:优化大肠杆菌XL1-blue菌株的电穿孔转化条件。方法:采用不同的条件组合转化大肠杆菌XL1-blue菌株,对转化条件进行优化。结果与讨论:用电穿孔方案转化大肠杆菌XL1-blue菌株,可得到明显高于其他转化方法的转化效率。外加电场强度和作用时间是决定转化效率的关键因素,其间存在一定的互补关系,转化效率和外源基因浓度之间呈线性关系,并与细菌浓度密切相关。
OBJECTIVE: To optimize the electroporation transformation conditions of E. coli XL1-blue strain. Methods: Different conditions were used to transform E. coli XL1-blue strains to optimize the transformation conditions. Results and Discussion: Transformation efficiency of E. coli XL1-blue strain by electroporation was significantly higher than that of other transformation methods. The applied electric field strength and action time are the key factors that determine the transformation efficiency. There is a certain complementary relationship between them. The linear relationship between the transformation efficiency and the exogenous gene concentration is closely related to the bacterial concentration.
