目的:观察七叶皂苷钠冻干粉对缺血性脑水肿小鼠的治疗作用及机制研究。方法:制备小鼠脑缺血模型,将实验小鼠随机分为4组,空白对照组、模型组、地塞米松组、七叶皂苷钠组。七叶皂苷钠组于尾静脉注射七叶皂苷钠3.6 mg/kg,地塞米松组按等浓度的地塞米松磷酸钠注射液尾静脉注射,模型组及空白对照组给予等容积的0.9%生理盐水,每天1次,连续给药7 d。干湿称重法测定各组大鼠脑组织含水量,ELISA法测定脑组织IL-1β和ET含量。结果:与空白组比较,模型组小鼠脑组织含水量显著升高,脑组织IL-1β和ET含量均升高,具有统计学意义(P<0.01);与模型组比较,地塞米松组、七叶皂苷钠组小鼠脑组织含水量降低,脑组织IL-1β和ET含量均降低,具有统计学意义(P<0.01)。结论:七叶皂苷钠冻干粉能减少缺血性脑水肿小鼠出血后脑水肿程度,改善脑组织病理变化,其作用机制与降低脑组织IL-1β和ET含量有关。
Objective: To observe the therapeutic effect of sodium aescinate lyophilized powder on ischemic cerebral edema in mice and its mechanism. Methods: The mouse model of cerebral ischemia was prepared. The experimental mice were randomly divided into 4 groups: blank control group, model group, dexamethasone group and sodium aescinate group. Sodium aescinate sodium group in the tail vein injection of sodium aescinate 3.6 mg / kg, dexamethasone group according to the same concentration of dexamethasone sodium phosphate injection tail vein injection, the model group and the blank control group were given 0.9% of the volume of physiology Saline, once daily, for 7 consecutive days. The water content of brain tissue in each group was measured by wet-dry weighing method. The contents of IL-1β and ET in brain tissue were determined by ELISA. Results: Compared with the blank group, the water content of brain tissue in model group increased significantly, and the levels of IL-1β and ET in brain tissue increased, with statistical significance (P <0.01). Compared with model group, dexamethasone group The content of brain water and the content of IL-1β and ET in brain of the mice in the sodium aescinate group decreased, with statistical significance (P <0.01). Conclusion: Sodium aescinate lyophilized powder can reduce the degree of cerebral edema after hemorrhagic cerebral ischemia in mice with ischemic cerebral edema and improve the pathological changes of brain tissue. Its mechanism may be related to the decrease of IL-1β and ET in brain tissue.
