背景:急性冠状动脉综合征(acutecoronarysyndrome,ACS)发生通常是由于动脉壁不稳定粥样斑块的破裂和血栓形成造成的,基质金属蛋白酶(MMPs)通过降解细胞外基质蛋白,在血管重构及斑块不稳定性中发挥重要作用。目的:探讨MMP-3和MMP-12基因启动子多态性与中国汉族人群中ACS危险性的可能关系。设计:以ACS患者为研究对象,健康人群为对照组的病例-对照研究。单位:一所大学医院的检验系和一所医学院的生化与分子生物学系。对象:研究对象为武警湖北总队医院、武汉大学中南医院和华中科技大学梨园医院2002-03/2003-12的门诊和住院患者,分为ACS组和正常对照组。ACS组103例,男73例,女30例,平均年龄(53±10)岁,包括急性心肌梗死47例、不稳定心绞痛56例;正常对照组为100例无血缘关系的健康个体(冠脉造影正常),男67例,女33例,平均年龄(52±12)岁;以上研究对象均为湖北地区汉族人,均自愿参加本研究。方法:采用多聚酶链反应-限制性片段长度多态性(PCR-RFLP)分析方法,分别检测了湖北地区汉族103例ACS患者及100例健康人的MMP-3基因-16125A/6A和MMP-12基因-82A/G多态性的基因型。主要观察指标:两组基因型和等位基因分布频率的比较。结果:ACS组MMP-3基因的5A/6A+5A/5A基因型频率及5A等位基因频率显著高于健康对照组(41.7%比24
BACKGROUND: The occurrence of acute coronary syndrome (ACS) is usually caused by rupture and thrombosis of unstable atherosclerotic plaque. Matrix metalloproteinases (MMPs), through degradation of extracellular matrix proteins, Plaque instability plays an important role. Objective: To investigate the possible relationship between MMP-3 and MMP-12 gene promoter polymorphism and ACS risk in Chinese Han population. Design: A case-control study of ACS patients and control subjects. PARTICIPANTS: The subjects were outpatients and inpatients from Armed Police Corps Hospital in Hubei Province, Zhongnan Hospital of Wuhan University and Liyuan Hospital of Huazhong University of Science and Technology between March 2002 and December 2003, which were divided into ACS group and normal control group. 103 ACS patients, 73 males and 30 females, with an average age of 53 ± 10 years, including 47 cases of acute myocardial infarction and 56 cases of unstable angina pectoris.
