采用多种层析方法从广东桑根心材的醇提取物中分离了8个已知化合物,经波谱分析手段结合文献分析方法将其分别鉴定为:桑辛素C(1),桑辛素M(2),二氢桑色素(3),氧化白藜芦醇(4),2,4-二羟基苯甲醛(5),伞形花内酯(6),胡萝卜苷(7)和3-羟基-2-哌啶酮(8)。化合物1,3~6和8首次从广东桑Morus atpropurpurea中分离,化合物8亦首次从桑属植物中分离。综合采用DPPH,MTT法测试了多酚类化合物1~4的抗氧化和神经保护作用,结果表明化合物1~4均能清除DPPH自由基,抑制L-谷氨酸钠诱导海马神经元细胞HT22毒性,具有抗氧化和神经保护作用。在20μM浓度下,化合物2的活性显著。
Eight known compounds were isolated from alcohol extracts of Morus alba L. by a variety of chromatographic methods and identified by spectral analysis and literature analysis as follows: (2), dihydrocyanin (3), resveratrol (4), 2,4-dihydroxybenzaldehyde (5), ursolactone (6) Hydroxy-2-piperidone (8). Compounds 1, 3 ~ 6 and 8 were isolated from Morus atpropurpurea for the first time. Compound 8 was also isolated from Morus for the first time. The antioxidant and neuroprotective effects of polyphenols 1 ~ 4 were tested by DPPH and MTT assay. The results showed that compounds 1 ~ 4 could scavenge DPPH free radical and inhibit L-glutamate-induced HT22 toxicity in hippocampal neurons , With anti-oxidant and neuroprotective effects. Compound 2 activity was significant at a concentration of 20 [mu] M.
