目的:研究姜黄素对长期摄入酒精和不同量蛋白质的大鼠胰腺腺泡细胞损伤的保护作用。方法:实验分为5组,即正常对照(正常饲养)、高蛋白、低蛋白、高蛋白+姜黄素、低蛋白+姜黄素(以25%酒精代替饮水自由饮用,高、低蛋白质占总热量供给的32%、6%,喂饲6个月)组。在光镜和电镜下观察大鼠胰腺腺泡细胞结构变化,用比色法检测胰腺组织匀浆淀粉酶和脂肪酶的含量,TUNEL法检测腺泡细胞凋亡情况,免疫组化检测胰腺组织切片中环氧化酶-2(COX-2)的变化。结果:与高、低蛋白组比较,高、低蛋白+姜黄素组大鼠胰腺腺泡细胞髓样结构减少,线粒体肿胀减轻;淀粉酶和脂肪酶含量均显著升高(P<0.05);胰腺腺泡细胞凋亡显著减少(P<0.05);COX-2的表达降低。结论:姜黄素可预防摄入酒精联合过高或过低蛋白质的大鼠胰腺腺泡细胞损伤,延缓酒精性胰腺损伤的进程。
Objective: To study the protective effect of curcumin on rat pancreatic acinar cell injury induced by long-term consumption of alcohol and different amounts of protein. Methods: The experiment was divided into five groups: normal control (normal feeding), high protein, low protein, high protein + curcumin and low protein + curcumin (25% alcohol instead of drinking water to drink freely, high and low protein in total calories Supply 32%, 6%, fed 6 months) group. The morphological changes of pancreatic acinar cells were observed under light microscope and electron microscope. Results: Compared with the high and low protein groups, the myeloid structure of pancreatic acinar cells in high and low protein + curcumin groups decreased and the mitochondrial swelling decreased. The contents of amylase and lipase were significantly increased (P <0.05) Acinar cell apoptosis decreased significantly (P <0.05); COX-2 expression decreased. CONCLUSION: Curcumin can prevent the injury of pancreatic acinar cells in rats with alcohol overload or excess protein, and delay the process of alcoholic pancreatic injury.
