目的:探讨肿瘤患者外周血T淋巴细胞间期核和分裂期染色体AgNORs的相关性及临床应用价值。方法:应用银染色技术及KL型图像分析系统,检测74例食管癌、68例原发性肝癌患者和40例对照组的T淋巴细胞间期核和分裂期染色体AgNORs,结果分别以银染核仁面积与细胞核面积的百分比值(I.S%)和AgNORs数/细胞表示。结果:①食管癌和原发性肝癌患者间期核AgNORs的I.S%(5.28±0.86和5.37±0.94)显著低于对照组(7.56±0.78)(P<0.01),分裂期染色体AgNORs数/细胞(5.64±0.45和5.75±0.49)也显著低于对照组(7.35±0.55)(P<0.01);②这两种指标在食管癌和原发性肝癌患者间均无显著性差异(P>0.05);③食管癌、原发性肝癌患者及对照组的这两种指标呈正相关,相关系数分别为0.82、0.80和0.87,均为P<0.001。结论:肿瘤患者外周血T淋巴细胞间期核AgNORs的I.S%和分裂期染色体AgNORs数/细胞具有相关性,二者均可作为肿瘤的辅助诊断指标,但前者能自动计数、检测方便,结果准确可靠,比后者更适合在临床推广应用。
Objective: To investigate the correlation between T lymphocyte interphase and mitotic AgNORs in peripheral blood of patients with cancer and its clinical value. Methods: AgNORs of T lymphocyte interphase and mitotic chromosomes in 74 cases of esophageal cancer, 68 cases of primary hepatocellular carcinoma and 40 cases of control were detected by silver staining technique and KL image analysis system. The results were analyzed by silver staining Percentage of kernel area to nucleus area (IS%) and AgNORs / cells. Results: (1) The IS% (5.28 ± 0.86 and 5.37 ± 0.94) of interphase nucleus AgNORs in esophageal cancer and primary liver cancer patients were significantly lower than those in control group (7.56 ± 0.78) (P <0.01). The numbers of AgNORs / (5.64 ± 0.45 and 5.75 ± 0.49) were also significantly lower than those in the control group (7.35 ± 0.55) (P <0.01); ② There was no significant difference between the two indexes in patients with esophageal cancer and primary liver cancer (P> 0.05 ); ③ There was a positive correlation between the two indexes of esophageal cancer, primary liver cancer and control group, the correlation coefficients were 0.82,0.80 and 0.87, respectively, P <0.001. CONCLUSIONS: There is a correlation between the IS% of AgNORs in peripheral T lymphocytes and the number of AgNORs / cell in the mitotic stage of T lymphocytes in peripheral blood of patients with cancer, both of which can be used as ancillary diagnostic indicators for cancer. However, the former can be automatically counted and tested conveniently with accurate results Reliable, more suitable for clinical application than the latter.
