目的:采用HPLC法测定自制牛黄渗透泵片中胆酸含量。方法:用冰醋酸溶解自制牛黄渗透泵片,经硫酸-水混合溶液70℃水浴处理后,采用HPLC法测定,色谱条件:十八烷基硅烷键合硅胶为填充剂,乙腈-0.15%磷酸水溶液(40∶60)为流动相,检测波长为387nm。结果:胆酸在48μg/mL~240μg/mL(r=0.999 2,n=3)浓度范围内呈良好线性关系,平均回收率为99.60%,RSD=0.87%。结论:该方法可用于测定牛黄渗透泵片中胆酸含量,快速简便,准确有效。
Objective: To determine the content of bile acids in homemade bezoar osmotic pump tablets by HPLC. METHODS: Self-made bezoar osmotic pump tablets were dissolved in glacial acetic acid and the mixture was subjected to water bath treatment at 70 ℃. The chromatographic conditions were as follows: octadecylsilane bonded silica gel as filler, acetonitrile-0.15% phosphoric acid aqueous solution (40:60) as the mobile phase, detection wavelength of 387nm. Results: There was a good linear relationship between the concentration of cholic acid and the concentration of 48μg / mL ~ 240μg / mL (r = 0.999 2, n = 3) with the average recovery of 99.60% and RSD of 0.87%. Conclusion: This method can be used to determine the content of bile acid in the bezoar osmotic pump tablets. It is fast, simple, accurate and effective.
