目的研究护肝清脂片药物血清对脂肪变性肝细胞的甘油三酯代谢以及氧化应激水平的影响。方法以不同浓度游离脂肪酸(free fatty acid,FFA)混合物(油酸和棕榈酸)孵育Hep G2 24h,建立并筛选最佳Hep G2脂肪变性模型,分别以大鼠空白血清和非诺贝特药物血清为对照,采用油红O染色、试剂盒检测等方法检测护肝清脂片药物血清对Hep G2脂肪变性细胞模型脂滴形态、甘油三酯含量以及氧化应激相关指标的变化。结果护肝清脂片药物血清明显减小Hep G2细胞内脂滴数量和大小,降低细胞内甘油三酯、活性氧和丙二醛含量,同时增加总抗氧化能力、超氧化物歧化酶活性和谷胱甘肽含量。结论护肝清脂片药物血清抑制游离脂肪酸诱导的Hep G2细胞脂肪变性,其可能与清除Hep G2细胞内活性氧自由基以及提高Hep G2细胞的抗氧化能力、减轻细胞氧化应激损害有关。
Objective To study the effects of Huganqingzhi Tablet serum on triglyceride metabolism and oxidative stress in steatosis hepatocytes. Methods Hep G2 cells were incubated with different concentrations of free fatty acid (FFA) mixture (oleic acid and palmitic acid) for 24 h. The optimal model of Hep G2 fatty degeneration was established and screened. The blank serum and fenofibrate serum As control. The changes of lipid droplet morphology, triglyceride content and related indicators of oxidative stress in Hep G2 fatty degeneration cell model were detected by oil red O staining and kit detection. Results Huganqingzhi tablet serum could obviously decrease the number and size of lipid droplets in Hep G2 cells, decrease the contents of triglyceride, reactive oxygen species and malondialdehyde in cells, meanwhile increase the total antioxidant capacity, superoxide dismutase activity and Glutathione content. Conclusion Huganqingzhi tablet can inhibit the fatty degeneration of Hep G2 cells induced by free fatty acids, which may be related to the removal of reactive oxygen species in Hep G2 cells and the anti-oxidative ability of Hep G2 cells and the reduction of cell oxidative stress.
