本文以检测K562细胞增殖活性为例,对MTT法的实验条件进行了探讨,结果表明:20%SDS~50%DMF作甲溶解缓冲液效果最佳,其溶解的甲产物吸收峰为570nm波长。测定细胞范围800~400000/孔,能广泛地应用于多种细胞存活和增殖的测定。
In this paper, the proliferative activity of K562 cells was detected as an example. The experimental conditions of MTT assay were discussed. The results showed that 20% SDS-50% DMF was the best for the chelation buffer, and the solubilization peak of the lysate was 570 nm. The measurement range of 800 to 400,000 cells/well can be widely applied to the determination of a variety of cell survival and proliferation.
