目的 应用Langendorff 模型研究缺血预处理(IPC) 对兔未成熟心肌缺血/ 再灌注损伤的影响并探讨其可能机制。方法 取3 ~4 周未成熟离体兔心24 只,随机分为IPC组和Con 组,根据缺血前IPC的次数将IPC组分为IPC1 、IPC2 和IPC3 3 个亚组,每组6 只。IPC1、IPC2 和IPC3 3 个亚组的心肌常温平衡灌注20 min 后分别给予1、2、3 个周期的5 min 缺血和5 min 再灌注,而Con 组心肌在缺血前平衡灌注20 min 后续灌30 min。所有心肌均常温缺血45 min,再灌注30 min。于平衡末,缺血前,再灌注后即刻、3、5、10、20、30 min 时测定心脏的功能指标以及再灌注末心肌组织中ATP、丙二醛(MDA) 含量及超氧化物歧化酶(SOD) 的活性。结果 IPC1、IPC2 组的LVDP、+ dp/dtmax 及- dp/dtmax的恢复百分比显著高于Con 组( P < 0.05),IPC2 组+ dp/dtmax 显著高于IPC3 组( P < 0 .05) 。IPC1、IPC2 组的LVEDP值显著低于Con 组( P< 0.05) 。IPC可维持心肌组织中的ATP在较高水平,以IPC2 组最
Objective To investigate the effect of ischemic preconditioning (IPC) on immature myocardial ischemia / reperfusion injury in rabbits and to explore its possible mechanism by Langendorff model. Methods Twenty-four immature rabbit hearts from 3 to 4 weeks were randomly divided into IPC group and Con group. IPC groups were divided into IPC1, IPC2 and IPC3 subgroups according to the number of IPC before ischemia, with 6 . IPC1, IPC2 and IPC3 three subgroups of normal myocardial perfusion after 20 min were given 1, 2 and 3 cycles of 5 min ischemia and 5 min reperfusion, and Con group myocardial perfusion before ischemia in 20 min continued Irrigation 30 min. All myocardial ischemia at room temperature 45 min, reperfusion 30 min.
