OBJECTIVE: To observe the effects of insulin-like growth factor I (IGF-I) on the proliferation of rat osteoblasts, the synthesis of alkaline phosphatase (ALP) and the formation of calcified nodules in vitro. METHODS: Cultured rat osteoblasts were stimulated with different concentrations of IGF-I, MTT assay was used to determine the cell proliferation. ALP activity in cell lysate was determined by p-nitrophenol phosphate assay. Calcification was measured by Von Kossa staining The number of sections. Results: IGF-I at a certain concentration significantly increased the number of osteoblasts in rats (P <0.05). The effect of IGF-I at a concentration of 0.1-100.0 ng / ml was positively correlated with the concentration of IGF-I. The ALP synthesis in osteocytes was significantly higher than that in the control group (P <0.05). The number of calcified nodules in osteoblasts stimulated by IGF-I was significantly higher than that in the control group (P <0.001). CONCLUSION: IGF-I can increase the number of osteoblasts cultured in vitro and promote ALP synthesis and calcification in osteoblasts, suggesting that IGF-I may directly promote bone formation.
