目的通过HPLC法测量清开灵颗粒中黄苷的含量。方法反相C18色谱柱,以甲醇-水-磷酸(47∶53∶0.2)为流动相,检测波长280nm,流速1.0ml/min,柱温30℃,进样量10μl。结果HPLC能够准确测量黄芩苷的含量,在10.54~120.18μg/ml范围内峰面积与浓度呈良好的线性关系(r=0.9996,n=5)。平均回收率为99.1%(RSD=0.45%,n=6)。结论HPLC法测量黄芩苷是一种方便快捷的检测方法,其结果准确可靠。
Objective To measure the content of xanthosine in Qingkailing Granules by HPLC. Methods Reversed phase C18 column with methanol-water-phosphoric acid (47∶53:0.2) as mobile phase, detection wavelength of 280 nm, flow rate of 1.0 ml/min, column temperature of 30°C, and injection volume of 10 μl. Results The content of baicalin could be accurately measured by HPLC. The linear relationship between peak area and concentration was in the range of 10.54-120.18 μg/ml (r=0.9996, n=5). The average recovery was 99.1% (RSD = 0.45%, n = 6). Conclusion The HPLC method for the determination of baicalin is a convenient and rapid method. The results are accurate and reliable.
