目的:筛选天麻素经鼻给药制剂的吸收促进剂种类和用量。方法:采用HPLC测定天麻素含量,流动相乙腈-0.05%磷酸(3∶97),检测波长220 nm。采用Franz水平扩散池,以家兔鼻黏膜为渗透屏障,比较冰片、丙二醇、水溶性氮酮对天麻素经鼻给药制剂的促渗透效果,确定吸收促进剂种类和用量。结果:3种吸收促进剂均对天麻素经鼻给药制剂有促渗作用,5.0%水溶性氮酮的促渗效果最好,3 h内天麻素累积渗透量达3 470.54μg·cm-2,增渗倍数1.922。结论:5%氮酮可作为天麻素经鼻给药制剂的吸收促进剂。
OBJECTIVE: To screen the types and amounts of absorption enhancers for nasal administration of gastrodin. Methods: The content of gastrodin was determined by HPLC. The mobile phase consisted of acetonitrile - 0.05% phosphoric acid (3:97) and the detection wavelength was 220 nm. Franz horizontal diffusion cell was used to improve the osmotic barrier of nasal mucosa in rabbits. The promoting effect of borneol, propylene glycol and water-soluble Azone on nasal administration of gastrodin was compared, and the types and dosage of absorption enhancer were determined. Results: The three kinds of absorption enhancers all had a promoting effect on nasal administration of gastrodin. The best effect of 5.0% water-soluble azone was 3 470.54 μg · cm-2 , Infiltration multiples of 1.922. CONCLUSIONS: 5% Azone can be used as an absorption enhancer for nasal administration of gastrodin.
