目的 研究 型胶原及其受体系统在不同代次成骨细胞内的表达情况。方法 选用原代、第 6代及第 15代人胚骨膜成骨细胞 ,S- P免疫组织化学染色观察 型胶原及整合素α2 β1 的表达情况 ,定量 RT- PCR技术检测 型胶原及整合素α2 β1 的 m RNA表达情况。结果 型胶原及其受体在不同代次成骨细胞内均有表达 ,免疫组织化学未发现各代次间表达量的差异 ;不同代次的人胚成骨细胞都有 型胶原及其受体 -整合素α2 β1 的m RNA表达 ,表达量随成骨细胞传代次数的增加逐渐减弱。结论 型胶原是通过整合素受体α2 β1 对成骨细胞发挥促分化和促粘附作用 ;该系统的表达减弱将导致成骨细胞功能衰退
Objective To study the expression of collagen and its receptor system in different generations of osteoblasts. Methods The primary, sixth and 15th generation human osteoblasts were used to detect the expression of collagen and integrin α2β1 by S-P immunohistochemistry. The expressions of collagen type Ⅱ and integrin α2 β1 m RNA expression. Results Collagen and its receptor were expressed in different generations of osteoblasts, and immunohistochemistry did not find any difference in expression between generations; in different generations of human embryonic osteoblasts, both type collagen and its receptor The m RNA expression of integrin α2 β1 decreased gradually with the passage of osteoblasts. Conclusions Collagen is an agent that promotes differentiation and adhesion through the integrin receptor α2β1; the decreased expression of this system leads to a decline in osteoblast function
