目的 :形态学观察雌激素受体基因敲除小鼠脾脏免疫学指标是否发生改变。方法 :应用免疫组织化学和免疫荧光法进行检测。结果 :雌激素受体基因敲除小鼠 ,尤其是ERβ敲除小鼠 (BERKO)的脾脏出现多种异常 ,如促炎症细胞因子和诱导型一氧化氮增高 ,脾细胞增殖增强并能抵抗雌激素的抑制作用 ,IgM /IgG表达也明显增高。雌激素受体缺乏小鼠脾脏NF kB的激活可能是脾细胞过分活跃的原因。结论 :雌激素受体 ,尤其是ERβ在介导雌激素对免疫反应的调节中起重要作用 ,ERβ缺失可能增加机体对自身免疫性疾病的敏感性
OBJECTIVE: To observe whether the spleen immunological parameters of estrogen receptor knockout mice have changed after morphological observation. Methods: Immunohistochemistry and immunofluorescence were used for detection. Results: The estrogen receptor knockout mice, especially the ERβ knockout mice (BERKO), showed various abnormalities in the spleen such as increased proinflammatory cytokines and inducible nitric oxide, enhanced splenocyte proliferation and resistance to estrogen Inhibition of hormones, IgM / IgG expression was also significantly higher. Estrogen Receptor Deficiencies Activation of NF kB in mouse spleens may be responsible for splenocyte hyperactivity. CONCLUSIONS: Estrogen receptors, especially ERβ, play an important role in the regulation of estrogen regulation of the immune response. ERβ deficiency may increase the body’s sensitivity to autoimmune diseases
